CADM1: A cell adhesion molecule closely related to the invasion and metastasis of cancer

Cancer cells restricted to the area where it occurred (primary tumors) can be effectively removed or killed through surgery or radiotherapy, but once it metastasizes, the treatment options are limited to chemotherapy or immunotherapy. The majority of deaths from solid tumors are caused by invasion or metastasis, and the medical community is seeking to develop drugs to inhibit invasion and metastasis of cancer.

In order to achieve this, scientists must first understand how metastasis occurs. Our group is studying this mechanism through a molecular biological approach.

Metastasis begins when adhesion between cancer cells is lost. We focused on the CADM1 gene, which is closely linked to the adhesion of the cells. CADM1 is mainly involved in cell adhesion in epithelial tissues such as the lungs, brain and testes, and the progression of cancers originating from epithelial cells, such as lung cancer, is frequently accompanied by a loss of or decrease in the expression of this gene. Furthermore, restoring CADM1 to the cancer cells inhibits their growth and metastasis. This indicates that the adhesion between cells mediated by CADM1 inhibits the growth, invasion and metastasis of cancer cells, making CADM1 a tumor suppressor gene.

However, CADM1 is overexpressed in cancers such as adult T-cell leukemia (ATL) and small cell lung cancer, where its promotion of adhesion to vascular endothelial cells causes the cancers to invade and metastasize. CADM1 therefore acts as an oncogene in this case.

Analyzing molecular interactions between cells to understand the dual roles of cell adhesion molecules

We believe that one of the reasons why CADM1 works in opposite ways depending on the tissue is because of the difference in the molecules that CADM1 binds to on the surface of the cell membrane. We are carrying out ongoing research to find out what kinds of molecular interactions are taking place between CADM1 and the molecules it binds to.

Unfortunately, these interactions are extremely weak, making it very difficult to detect them using conventional molecular biological methods. To solve this problem, we recently began working with an SPRi (surface plasmon resonance imaging) device called the HORIBA OpenPlex.

We are using the OpenPlex to gather data on various parameters related to association and dissociation between CADM1 and the cell surface molecules of different types of tissue. We then identify the interactions and analyze factors such as binding affinity to study how the invasion and metastasis of cancer is controlled by cell adhesion.

If we can understand how CADM1 functions both as a tumor suppressor gene and as an oncogene, we expect that this will lead to the development of revolutionary new treatments to inhibit the invasion and metastasis of cancer. Most current molecular target drugs directly block the growth of cells, and drugs to inhibit invasion and metastasis have not yet been developed. We aim to build on our basic research to create completely new diagnosis and treatment methods that can control the invasion and metastasis of cancer.
